L Latar Belakang : Teknik home bleaching menggunakan carbamide peroxide sering digunakan karena tingkat kesuksesannya yang tinggi dan efek samping yang sedikit. Meskipun prosedur ini relatif aman, namun dapat menyebabkan perubahan struktur permukaan email gigi dan mempengaruhi perlekatan bakteri Streptococcus mutans pada permukaan email gigi. Tujuan : Penelitian ini bertujuan untuk mengetahui pengaruh carbamide peroxide terhadap perubahan struktur permukaan email gigi dan perlekatan Streptococcus mutans. Metode : Sampel dikelompokkan menjadi 4 yaitu, kelompok kontrol, kelompok sampel dengan aplikasi carbamide peroxide konsentrasi 10%, 15%, dan 35%. Uji perubahan struktur permukaan dilakukan secara subjektif berdasarkan pengamatan menggunakan mikroskop stereo dengan perbesaran 40x pada permukaan bukal gigi premolar 1 rahang atas sebelum dan sesudah perlakuan. Sampel direndam dalam kultur Streptococcus mutans dan diinkubasi selama 24 jam. Selanjutnya, sampel dibilas dengan Phosphate Buffered Saline dan dilepaskan perlekatannya. Uji perlekatan bakteri dilakukan dengan metode turbidimetri dan Total Plate Count. Hasil : Perubahan struktur permukaan email gigi sangat terlihat pada kelompok dengan konsentrasi 35%. Hasil uji ANOVA 1 jalur menunjukkan adanya perbedaan signifikan (p<0,05) pada uji perlekatan bakteri. Jumlah bakteri terbanyak ditemukan pada sampel yang diaplikasikan carbamide peroxide konsentrasi 35% sebanyak 3.034.000 CFU/mL (OD 0,06). Kesimpulan : Carbamide peroxide mempengaruhi perubahan struktur permukaan email dan perlekatan Streptococcus mutans.

Even though this procedure is relatively safe, but it can cause the changes of enamel surface structures and influence Streptococcus mutans’ adherence. Objective : The aim of this study is to investigate the influences of carbamide peroxide to enamel surface structures and streptococcus mutans adherence. Method : The samples are divided into 4, namely control group, sample group with application carbamide peroxide 10%, 15%, and 35%. The changes of surface structures test is done subjectively based on observation using microscope stereo with 40x magnified on the buccal surface of first upper premolar before and after bleaching treatments. Samples were immersed in Streptococcus mutans culture and incubated for 24 hour. Furthermore, the samples is rinsed using PBS and the bacterial attachment is released. Bacterial adherence is tested using Turbidimetry and Total Plate Count methods. Result : Carbamide peroxide can change enamel surface structures especially on 35% concentration. One way ANOVA test result showed there was significant difference (p < 0,05) on bacterial adherence test. The highest number of bacteria was found on samples that were applied with 35% carbamide peroxide as much as 3.034.000 CFU/mL (OD 0,06). Conclusion : Carbamide peroxide can change enamel surface structures and Streptococcus mutans’ adherence.
